These are the basic laboratory sample processing steps.

Bead-beating

Samples are bead-beaten to ensure tissues and cells are degraded before DNA extraction

DNA extraction

DNA is extracted using chemical and bead-based extraction.

DNA shearing

DNA is shared by sonication to the appropriate fragment size for short-read sequencing

Library building

Sequencing adaptors are attached to DNA molecules using ligation and sequencing indices through PCR

5

Sequencing

Multiple samples are sequenced in each sequencing lane aiming at 5x genome coverage and 8GB of metagenomic DNA
